Introduction

 Research is conducted to construct evidence connected to the development of cancer prevention by clarifying the causes of cancer in humans by leveraging large-scale cohort studies and others of local residents.

Research Activities

1)  Utilization of Biorepository Samples for the Epidemiological Research

 We initiated a new case-cohort study based on a five-year follow-up survey conducted as part of the Japan Public Health Center-based prospective study (the JPHC study) between 1995 and 1998 to further advance plasma biomarker research. The base cohort consisted of 32,706 individuals who provided plasma samples and answered questionnaires during the survey. Within this base cohort, 5,377 cancer cases were identified during approximately 20 years of follow-up, and 5,500 individuals were randomly selected as a sub-cohort. A total of 9,972 plasma samples were prepared for future research as 905 cancer cases were documented within the sub-cohort.

2)  Epidemiological Research on Cancer Etiology and Prevention

 We have conducted molecular epidemiological research to investigate cancer causes and prevention, utilizing biological samples collected in the JPHC study. In 2022, we published a study in an international journal that examined the association between pre-diagnostic iron-related biomarkers and cancer incidence. One significant finding was that individuals with iron overload had an approximately 4.5-fold higher risk of developing liver cancer compared to those with normal iron levels. In another international journal publication, we reported that the cancer-promoting effect of the Arg72Pro polymorphism of the TP53 gene was more pronounced in individuals with BMI <25 kg/m². Additionally, we co-authored several manuscripts on genetic research as members of the international consortium for breast and colorectal cancer.

3)  Consortium on Large-scale Molecular Epidemiological Studies in Japan

 By utilizing the Japanese Consortium of Genetic Epidemiology, we have conducted several Mendelian randomization analyses. In 2022, we published our findings in an international journal, revealing a statistically significant positive association between genetically predicted total cholesterol levels and the risk of colorectal cancer. In another international journal publication, we reported that there was no statistically significant association between genetically predicted blood vitamin D levels and the risk of overall cancer or colorectal cancer.

Education

 We supervised the research of four graduate students.

Future Prospects

 By constructing and utilizing research infrastructure for epidemiological studies, we hope to contribute to developing evidence for personalized cancer prevention and promoting a lifetime of health.
